摘要
The last step of aldosterone (ALDO) biosynthesis consists in the cytochrome P450-mediated mitochondrial oxidation of 11-deoxycorticosterone (DOG) through several pathways starting at its metabolites corticosterone (B) and 18-hydroxy-11-deoxycorticosterone (18OHDOC). For the last 30 years, investigations were undertaken on the number and identity of such cytochrome P450 enzymes. Actually the following model is accepted: in bovine adrenal gland cytochrome P450 11 beta/18-hydroxylase/aldo synthase catalyzes the whole reaction (from DOC to ALDO) being present in the entire adrenal cortex. By contrast, in rat, mouse and human adrenal gland the whole reaction is carried out by cytochrome P45011B12, only present in zona glomerulosa, while cytocrome P45011B1 catalyzes the conversion of DOC into B or 18OHDOC in zona fasciculata. Although this model is supported by cellular and molecular biochemistry experiments, it does not explain some experimental facts.
