High-level production and characterization of lactase from a newly isolated fungus Trametes sp. LS-10C

摘要
An efficient lactase production fungus Trametes sp. LS-10C was isolated and identified, and the lactase producing medium was optimized by response surface methodology (RSM) in shake flask fermentation. The highest lactase activity obtained at 9 d in shake flask and 10-L fermentator were 588.61 +/- 2.2 U mL(-1) and 873.82 +/- 22.91 U mL(-1), respectively. The molecular weight of purified lactase determined by SDS-PAGE was around 65 kDa. The optimal pH was 4.0-4.5 and the optimal temperature was 40 degrees C. Among the tested substrates of ABTS, DMP and guaiacol, both the highest values of V-max (11.4 +/- 1.5 mM min(-1)) and k(cat)/K-m, (6653.2 +/- 18.2 mM(-1) s(-1)) were found for ABTS and the lowest K-m value (38.1 +/- 1.7 mu m) was found for DMP.
