摘要：
DNA sequencing is a widely used technique; unfortunately, traditional methods do not apply to large DNA fragments lacking usable restriction sites. By combining both "star activity" of restriction endonucleases and template-independent activity of Taq polymerase, we developed a simple and reproducible method to overcome this difficulty.
