THE KINETICS OF AMPHOTERICIN-B INTERACTION WITH THE MUSCLE-FIBER MEMBRANE

摘要
We investigated the amphotericin B (10(-6)-10(-5) M) induced catio conductance in frog skeletal muscle fibres. The kinetics of the conductance decrease after amphotericin B was removed from the bathing solution consisted of two parts: the first with the relaxation rate constant rho = 2.03 +/- 0.21 min-1 and the slower second part depending on the amphotericin concentration. The value of rho calculated from the conductance rise was smaller than that for the conductance relaxation kinetics in the same experiment. The relaxation rate constant declined with temperature (Q10 = 2.8) and increased in prolonged experiments (greater-than-or-equal-to 80 min) after the repeated application of amphotericin B. The possible role of the nonconductive forms of amphotericin B interaction with the muscle fibre membrane is discussed.
