摘要
The unfolding of Trichosanthin (TCS) under acidic pH and guanidine chloride (GuHCl) has been investigated by H-1 NMR methods. The variation of C2H chemical shift by pH and GuHCl titration indicates that His(51) locates in the hydrophobic core of the large domain of TCS and an intermediate state is proposed during the acidic unfolding process. In alkaline condition (pH 12.0), It is also observed by 2D NOESY that the large domain still contains hydrophobic region while the small domain is random.
