Aberrant renal expression of peroxisome coactivator PGC-1 alpha and its regulators (Sirt1 & GSK3 beta) in rats with diabetic nephropathy

摘要
The dysregulation of peroxisome proliferator-activated receptor-gamma coactivator-1 alpha (PGC-1 alpha) has been implicated in diabetes mellitus. However, the role of PGC-1 alpha in diabetic nephropathy (DN) is still inconclusive. Our aim was to evaluate the renal expression of PGC-1 alpha and its regulators: silent information regulator 2 homolog (Sirt-1) and glycogen synthase kinase3 beta (GSK3 beta) in the diabetic rats. Nineteen adult male rats were randomly divided into the control group (n = 6) and the diabetic group (n. = 13). The expression of these genes was performed by quantitative real-time PCR (qRT-PCR) in the renal tissues. Also, catalase activity was measured by a colorimetric method. The diabetic rats had higher expression levels of PGC-1 alpha and GSK3 beta; however, the expression of Sirt-1 and catalase activity was significantly lower in comparison to the control group (p < 0.05). We concluded that the aberrant expression of PGC-1 alpha, Sirt-1, and GSK3 beta may be associated with the developing of DN.
