Equations have been developed which describe the effect of magnesium ion concentration and total magnesium concentration on the apparent equilibrium constant of the reaction catalysed by aconitase. A computer procedure is described which selects values of substrate‐magnesium stability constants which give the closest fit of the equations to experimental points. Values are given of apparent stability constants of magnesium citrate and isocitrate complexes, calculated in this way from equilibrium data obtained under a variety of conditions. The use of the aconitase equilibrium to measure magnesium ion concentration is shown to suffer from sensitivity to ionic strength. Copyright © 1969, Wiley Blackwell. All rights reserved
