Recent advances in the use of fluorimetric derivatization techniques in association with high-performance liquid chromatography (HPLC) for analytical purposes are presented. Formation of derivatives prior to chromatography is useful for both HPLC as well as thin-layer chromatography (TLC). Post-column reaction detection in HPLC may be likened to TLC post-chromatographic spray techniques albeit with important differences. Fluorescent derivatives produced from phenols, amines, carboxylic acids, aldehydes, ketones, and others are often sensitive enough to be detected in low nanogram quantities or less by HPLC.
