DNA replication in eukaryotic cells is initiated at sites in the DNA known as origins. Studies in yeast have identified a number of the genes and proteins that may be involved in this process. In this review, we concentrate largely on those genes or proteins that are required for initiation of DNA replication and for which there is some evidence for a role at origins.
