Immunoadsorption chromatography has been used to isolate pig-specific soluble muscle proteins from crude pig protein extracts. Pig-specific polyclonal antibodies against soluble pig muscle proteins immobilised on a Protein A-Sepharose CL-4B matrix were used to adsorb the corresponding antigens. Anlysis of the crude soluble pig muscle proteins by sodium dodecylsulphate polyacrylamide gel electrophoresis (SDS-PAGE) showed the existence of 12 protein subunits in the gel. SDS-PAGE results of the affinity chromatography recovered proteins, showed 7 protein subunits, two of them being enriched after the immunopurification stage. These affinity-recovered pig-specific soluble muscle proteins may be used as antigens in the development of monoclonial antibodies to detect and quantify the presence of pig meat in meat mixtures.
