DETECTION OF PRUNE DWARF VIRUS AND PRUNUS NECROTIC RINGSPOT VIRUS BY MEANS OF A RAPID VARIANT OF DOT-IMMUNOBINDING ASSAY

First results are presented regarding the detection of prune dwarf virus (PDV) and Prunus necrotic ringspot virus (PNRV) in dormant buds of plum and sour cherry trees by means of a Dot-Immunobinding Assay (DIA). The assay is based on the SERION(R) IMMUNO TAB produced by the VIRION/Wurzburg institute. The results obtained from a simultaneous variant of DAS-ELISA are used to rate the reliability of detection. Several modifications of DIA were examined which allowed to shorten the test time to 65 minutes (PDV) and 85 minutes (PNRV), respectively. The test system is reliable even a temperatures below 37-degrees-C. Therefore, the way is open to rapid diagnosis in open ground in order to solve specific problems.
