摘要
The mechanism of the H-phosphonate internucleotide condensation in the presence of different bases has been investigated by means of P-31 NMR spectroscopy and kinetic studies. Pyridine and quinoline are shown to exert nucleophilic catalysis at the step of the phosphorylation of nucleoside component by mixed anhydride. The activation of P-component by pivaloyl chloride in the presence of more powerful nucleophylic catalysts, such as Melm, DMAP, pyridine N-oxides, lead to rapid formation of acylphosphonate-base complexes, which display low phosphorylating activity.
