EXAMINATION OF POLY(U)-DEPENDENT BINDING OF TRNA(PHE) TO RIBOSOMAL P-SITE BY NITROSOETHYLUREA MODIFICATION

摘要
A study was made of nitrosoethylurea alkylation of tRNA(Phe) phosphates bound to the P site of E. coli 70S ribosomes and 30S subunits. Upon binding of tRNA to the 30S subunit, the reagent was not attached to nucleotides 23-44, i.e., to the anticodon loop and the neighboring region of the D stem. Upon binding to the P site of 70S ribosomes, nucleotides 45-49, 51, 54-55, 58-61, and 63 (T loop) were protected. The results obtained indicate that nitrosoethylurea can be used for studying the tRNA-ribosome interaction.
