A novel miniature Circulating, Long-Optical-path, Spectrof-lectrochemical Thin-layer (CLOSET) cell is described, which is particularly well-suited for the study of dilute redox enzymes, for the study of weak optical absorbers. Formal reduction potentials and n-values determined potentiometrically for the redox enzyme, cytochrome c, the weak optical absorber, Fe(CN)63- agree well with literature values. The CLOSET ceil operates as a miniature (600 μL), self-contained, magnetically driven, anaerobic recirculating pump, which couples a thinlayer electrochemical cell to a remote optical path of 1-cm length. The CLOSET approach enables dramatic reduction in electrolysis times relative to conventional bulk electrolysis cells of similar volume, dramatic increases In ptical sensitivity relative to optically transparent thin-layer electrochemical (OTTLE) cells. © 1979, American Chemical Society.
