Mass spectrometric sequence analyses of peptides can be accomplished in the following manner: at first the peptide is benzylated, during this process the sensitive SH-groups of cysteine are transformed to stable S-benzyl derivatives. Then a padiel hydrolysis is done. The peptide fragments are easily converted into volatile TFA-peptide methylesters. The small fragments are mass spectrometrically identified after gas chromatographic separation. The higher peptides may be separated by means of liquid chromatography; after that they likewise are identified by mass spectrometry. © 1969, Oxford University Press. All rights reserved.
