SYNTHESIS OF (+/-)-N-PHENETHYLMORPHINAN-6-ONE

摘要
The synthesis of (+)-N-phenethylmorphinan-6-one is synthesized first time using the intermediate (+)-2-(l-phenyl-lH-tetrazol-5yl) oxy] morphinan 6-one, which on acylation with acetic anhydride gave (+) -2- [( 1 -phenyl- 1H-tetrazol-5yl) oxy] -N-acylmorphinan-6-one. (+) -N-Acylmorphi-nan-6-one was obtained by catalytic hydrogenolysis of (+)-2-[(l-phenyl-lH-tetrazol-5yl) oxy] -N-acylmorphinan-6-one over Pd/C. On hydrolysis of (+)-N-acylmorphinan-6-one using methanolic sodium hydroxide afforded (+)-6-ketomor-phinan, which on alkylation with phenethyl bromide in presence of sodium bicarbonate gave the required compound, (+)-N-phenethylmorphinan-6-one, in 40% overall yield.
